Molecular Biology and Cell Culture Lab:
1. MTT Assay for cell proliferation/cytotoxicity
2. Cell culture techniques, passaging and maintenance of human cancer cell line
3. Media preparation and counting of cells
4. Cryopreservation: Freezing & thawing of human cancer cell line
5. Genomic DNA isolation from cancer cell line,
6. Genomic DNA estimation
7. Genomic DNA agarose gel electrophoresis
8. Total RNA extraction from human cancer cell line
9. cDNA synthesis from RNA (Reverse Transcription),
10. Polymerase Chain Reaction from cDNA (Reverse Transcription-PCR)
11. Agarose gel electrophoresis of PCR product
12. Total protein extraction from human cancer cell line
13. Protein estimation
14. SDS-PAGE electrophoresis of extracted proteins
15. Coomassie blue staining of SDS-PAGE gel
16. Induction of proliferation and differentiation in human cancer cell line
17. Cytospin slide preparation of hematopoietic cells
18. May-Gruenwald Giemsa Staining of cytospin slides of hematopoietic cells and visualizing morphology of differentiating cells
19. Plasmid DNA isolation
20. Plasmid DNA gel electrophoresis
21. Plasmid DNA gel extraction
22. Restriction digestion
23. Ligation
24. Agarose gel electrophoresis of restriction digested and ligated DNA
25. Preparation of competent cells
26. Transformation and plating on LBA plates

Bioinformatics tools for the above techniques:

28. Sequence Analysis: Pairwise sequence alignment, Multiple sequence alignment, BLAST tools
29. Designing of PCR primers,
30. Restriction analysis, restriction maps, plasmid maps,
31. Searching for homologous and paralogous sequences,
32. Promoter analysis, Transcription factor databases,
33. Whole genome data visualisation: browsing genomic data with the help of NCBI Genome viewer or UCSC Genome browser.

This program is designed for beginners of all ages who wants to become confidant in lab techniques. This is a practical hands-on workshop to train the candidates in animal cell culture, molecular biology techniques and MTT assay using the human embryonic and cancer cell lines for various experiments. It is supported by lectures on cell culture techniques, molecular biology assays, MTT assay, gene/protein expression/interaction networks in hematopoietic stem cells (HSCs) and cancer cells.”
